چکیده

One of serious obstacles of the aortopathies research is a considerable shortage of human aortic smooth muscle cells (SMC), which can be used to model the disease. SMC in most cases come from the whole aorta of transplant donors, which are rather difficult to access. In the course of coronary artery bypass graft (CABG) surgery a fragment of aortic tissue is excised to make a bypass root. In this study we show a possibility to use CABG leftover fragments of thoracic aorta as a source of human SMC for in vitro research. We isolated SMC from the fragments of aortic tissues obtained during CABG procedure and compared these cells to the cells that were isolated from aortic tissue of transplant donors. The content of key SMC contractile markers (SMA, SM22α, vimentin) as well as proliferation and migration rates, MMP-2 and MMP-9 activities were similar in CABG-derived SMC and in transplant donor derived SMC. In conclusion, leftovers of ascending thoracic aorta obtained during coronary artery bypass graft can be used as a source of human aortic smooth muscle cells for in vitro research.

Evaluation of Factors Influencing Liver Function Test in On-Pump Coronary Artery Bypass Graft Surgery

Background: Liver dysfunction during on-pump coronary artery bypass graft surgery (CABG) is a rare complication but is associated with significant morbidity and mortality. The ability to identify high-risk patients may be helpful in planning appropriate management strategies. We aimed to evaluate the factors influencing liver function tests during on-pump CABG.Methods: In 146 patients scheduled...

Calcium channel blockers enhance cholesteryl ester hydrolysis and decrease total cholesterol accumulation in human aortic tissue.

Calcium channel blockers (CCBs), which are used clinically for treatment of angina and hypertension, are known to inhibit calcium influx into arterial smooth muscle cells and thereby decrease smooth muscle cell contraction. In addition, they prevent cholesteryl ester (CE) accumulation, the hallmark of human atherosclerosis, in arteries of cholesterol-fed animals by cellular mechanisms that rema...

Diabetes mellitus induces hyperreactivity of 5-hydroxytryptamine (5-HT)-induced constriction in human internal thoracic artery and is associated with increase in the membrane protein level of 5-HT2A receptor.

Studies indicate that 5-hydroxytryptamine (5-HT) released from activated platelets in coronary artery bypass grafting (CABG) induces 5-HT2A receptor-mediated graft spasm. We previously reported that 5-HT-induced constriction of human endothelium-denuded saphenous vein (SV) was significantly augmented in patients with diabetes mellitus (DM) than in patients without DM (non-DM), without changes i...

Predictors' issues of intubation time after coronary artery bypass graft surgery

Abstract Introduction: Cardiovascular illness is the common disease in developed countries. Among of these illnesses the coronary artery failure significantly counted as a high rate of heart disease in the world. The main treatment for this condition is replacing coronary artery bypass graft (CABG) heart pump under surgical crew including skilled nurses. Objective: This study aimed to det...

Introduction: Genetically presence or absence of one series of carbohydrate molecules (as an agglutinogen) on the red blood cells membranes are determine ABO blood group systems. Studies showed that ABO blood groups are involved in pathogenesis of one human disorders set such as cardiovascular diseases. Thus, in this study, the prevalence rates of cardiovascular diseases were assessed in AB...
